Rafael Borre caused the jam-packed Frankfurt end to explode 12 minutes later by pouncing from close-range to equalise.
That sent the game to extra-time, and with neither team able to make a further breakthrough, that meant it would come down to spot-kicks to decide it.
James Tavernier, Steven Davis and Scott Arfield had slotted confidently from 12 yards for the Gers.
The Germans also scored their first three as well before Ramsey stepped up.
The Welshman, who arrived from Juventus in January, had only been on the pitch a matter of seconds after coming off the bench late in extra-time.
But the former Arsenal man smashed it low and hard down the middle, allowing Kevin Trapp to save.
Filip Kostic put Frankfurt ahead, and even though Kemar Roofe converted again for Rangers, Borre stepped up to win it for Frankfurt.
Naturally, Ramsey looked devastated as the Bundesliga club celebrated wildly, with a number of his team-mates consoling him.

And he could barely look at the trophy as he walked across the podium past it to collect his runners-up medal.
Now he might be best advised to stay off social media for a few days as a number of supporters quickly began trolling the 31-year-old.
One wrote: “Aaron Ramsey left Arsenal but Arsenal didn’t leave Aaron Ramsey.”
Another tweeted: “The Arsenal bottle job DNA is still in Aaron Ramsey’s veins unfortunately.”
A fellow tweeter wrote: “Aaron Ramsey really has that Arsenal bottling blood running through his veins.”
Meanwhile, Sun columnist and Arsenal supporter Piers Morgan commented: “Sympathies with @aaronramsey – can’t be anything worse in football than missing the only penalty to cost your team a European trophy. Feel for him.”
Rangers legend Ally McCoist told BT Sport: “You’ve got to feel for Aaron Ramsey. Anybody can miss a penalty.”
However, former Scottish international Pat Nevin was less forgiving on BBC 5Live, saying: “Torture. Every time you lose a cup final is torture. After the work they put in, after the fact they got better as the game went on.
“The standard of penalties was astonishingly high except for Ramsey. He has to live with that for the rest of his life.
“What an opportunity Rangers had. They’ll kick themselves. It was there to be won.”
