But he will now be replaced by Don Robertson, who was initially supposed to be the fourth official.
Collum suffered a calf injury in his warm up and had to pull himself out less than ten minutes before the match was due to start.
The 44-year-old was named as the referee for the cup clash last week.
Breaking the news on Viaplay, broadcaster Emma Dodds said: “We are always first with team news on Viaplay Sport and we’ve got some more breaking news coming to you from this game.
“Willie Collum is no longer the referee in this one. He has been injured in the warm-up with a calf strain so Don Robertson who was the fourth official is now taking charge.”
Collum has refereed four Scottish Cup finals in his time so may yet be given the nod for the showpiece in June.
He was the whistler last season when Rangers won the trophy with a 2-0 extra time win over Hearts, though, so duties may be given to someone else rather than having the same referee for two years in succession.
Before that he officiated the 2018/19 finale between Hearts and Celtic, the 2014/15 fixture between Inverness and Celtic and the 2012/13 event when Hibs faced Celtic.
